Check out what's under construction, when. You might need to take an alternate route depending on how the construction plays out. And try not to get stuck on 28th or 30th at rush hour, because it's going to take forever to get where you're going. There are also traffic cameras set up, so if you run the red light you will get your picture taken and get a nice fine in the mail.
Try auto trader or other online search engines, or just browse. There's a great dealership on 30th and pearl called Pollard Friendly Motors, I bought my Audi there a couple of years ago. It seems like most people buy or lease, I don't know many who have purchased cars directly from sellers on Craigslist. Make sure to get a carfax, Boulder is a really active town and someone may have taken their car up 70 to ski in the Mountains and gotten in an accident!
Driving in Boulder is really interesting, because it can go from congested nightmare to pleasant country drive really quickly. My favorite thing to do is drive on South Boulder Road from Boulder to Lafayette on a summer night. I remember doing this one time a couple years ago, and there was a lightning storm on the horizon. As I drove East, I could see the purple lightning lighting up the sky. There were fields all around me, I was nowhere near a highway, there was no traffic, and the entire road was mine. If you time it right you can hit every green light going East. Sometimes you'll also see wildlife - the other morning when I was driving to work I saw a deer and her fawn walking down the culvert on the side of the road.
South Boulder Road. South Boulder Road is great because it gets me to where I want to go, without having to drive on Highway 36 (which seems to always be under construction). I especially like the stretch between Cherryvale and McCaslin - there aren't a lot of lights, and there's a hill to drive up (or down). There's a winery going in, and a monastery, which is very Boulder! There isn't a lot of development around South Boulder road, it's really just for driving, which is nice.

Be extremely cautious and aware of bicyclists and pedestrians. While Boulder is plenty car friendly, it is also heavily laden with people who utilize other forms of transportation, bicycles in particular. For people who aren't familiar with this many non-car travelers being on the road, it can be a bit of a shock, or easy to get distracted and forget to be mindful of them.
There are several dealerships in Boulder, ranging from pre-owned/used/discount dealerships to higher end ones. 28Th Street alone, the road mentioned previously, has a big line-up of dealerships that span this spectrum.
My favorite memory of driving in Boulder is from 5 years ago, my freshman year of college. It was a Saturday afternoon in September, the day of the first CU football game. My best friend Taylor and I had driven over to Pearl Street for lunch and to walk around, and after finishing we got in the car and drove back up "the Hill" (Broadway St) and were just cruising by the houses right near campus. Because it was the day of the football game and the weather was nice, everyone was out tailgating, having a good time on the lawns or on balconies. I remember we were both wearing CU gear, and every time we'd drive past a house with people partying out front, we would slow down and wave, make noise, etc. It was such a fun Saturday.
28Th Street. I like driving on 28th Street because of just how many things are accessible on it, and even when there's traffic it remains an easy, drivable road. The stretch from Colorado, right when it turns into 28th after being Highway 36, up through Pearl Street, even up to Valmont, is my favorite. There are so many opportunities to just make a quick turn and have access to restaurants, stores, and just fun places, like the 29th Street Mall. It helps that this road also turns into 36 when you want to leave the city, making it easy to cruise through Boulder before hopping on the highway and heading out of town.
